18 år efter Brødrenes legendariske "Mission for Gud", er det lykkedes Elwood Blues at slippe ud af fængslet, hvor han opdager, at meget har ændret sig i den tid, han har været væk! Hans partner, Jake, er forsvundet, hans band er gået i opløsning, og børnehjemmet, hvor han voksede op, er blevet revet ned. Elwood indser hurtigt, at han må drage ud på en helt ny mission: at få samlet det gamle band igen denne gang med hjælp fra en bluesfyldt bartender, deltage i Queen Mousettes "Battle of the Bands" og få en forældreløs dreng ind på rette spor igen. Midt i al forvirringen er han også nødt til at overbevise politiet om, at der er magi i musikken...
